چکیده

Ectodermal dysplasia (ED) is a group of syndromes characterized by abnormalities of structures of ectodermal origin. Up to now more than 150 different syndromes are known. Affected ectodermal structures are mainly hair, teeth, nails and sweat glands. ED can be classified by mode of inheritance (autosomal dominant, autosomal recessive and X-linked), by additional clinical symptoms (e.g. facial abnormalities) or by the structures involved. Hypohidrotic or “classical” ED (HED) is caused by mutations in the genes EDA, EDAR and EDARADD. Mutations in the X-linked EDA gene underlie most cases. EDAR and EDARADD are known to be associated with both autosomal dominant and autosomal recessive forms of HED. As many ED forms show overlapping clinical phenotypes, often many genes have to be analyzed to identify the causative mutation in a patient, therefore Sanger sequencing frequently takes several months and is laborious and cost-intensive. We have designed an accurate and fast molecular test for the 6 most common ED genes using next-generation sequencing. We developed a novel PCR-based protocol on a Roche 454 Junior platform for mutation screening of the genes EDA, EDAR, EDARADD, TP63, GJB6 and WNT10A. The verification and validation process was finished in October 2011, and the “ED panel” is now the routine diagnostic test for ED in our institute. During the last months several mutations in different genes could be identified with this method. A definitive diagnosis could be made rapidly in a number of so far „unsolved cases”. In summary, the next generation sequencing approach has the potential to increase the diagnostic sensitivity by expanding the number of genes that can be screened in parallel while also reducing expenditure of time and costs.

Comprehensive Analysis of Deafness Genes in Families with Autosomal Recessive Nonsyndromic Hearing Loss

Comprehensive genetic testing has the potential to become the standard of care for individuals with hearing loss. In this study, we investigated the genetic etiology of autosomal recessive nonsyndromic hearing loss (ARNSHL) in a Turkish cohort including individuals with cochlear implant, who had a pedigree suggestive of an autosomal recessive inheritance. A workflow including prescreening of GJ...

Panel-based testing for inherited colorectal cancer: a descriptive study of clinical testing performed by a US laboratory

Next-generation sequencing enables testing for multiple genes simultaneously ('panel-based testing') as opposed to sequential testing for one inherited condition at a time ('syndrome-based testing'). This study presents results from patients who underwent hereditary colorectal cancer (CRC) panel-based testing ('ColoNext(™) '). De-identified data from a clinical testing laboratory were used to c...

Advantages and Some Remaining Challenges in Hereditary Gastrointestinal Cancer Panel Testing

Colorectal cancer affects 1 in 20 men and women in their lifetime. About 30% of these cases have been shown to be familial while only about 5% are associated with a highly penetrant hereditary colon cancer syndrome. In many familial cases, however, no mutation in the commonly implicated CRC genes is found. With the development of next-generation sequencing, testing laboratories are now able to ...

Targeted next-generation sequencing appoints c16orf57 as clericuzio-type poikiloderma with neutropenia gene.

Next-generation sequencing is a straightforward tool for the identification of disease genes in extended genomic regions. Autozygosity mapping was performed on a five-generation inbred Italian family with three siblings affected with Clericuzio-type poikiloderma with neutropenia (PN [MIM %604173]), a rare autosomal-recessive genodermatosis characterised by poikiloderma, pachyonychia, and chroni...
